Molecular Customs of Several Dates Genotypes (Phoenix Dactylifera L.) Using Primary Rapd

Dates (Phoenix Dactylifera L.) are one of the most widely cultivated plants in various countries, particularly in the Middle East. The date palm is a vital crop that provides food, income, and employment for millions of people worldwide. However, the genetic diversity of dates is a significant concern, as it affects the crop's adaptability to environmental changes and its ability to produce high-quality fruits. In this study, we aimed to understand the molecular performance of several dates genotypes using random amplified polymorphic DNA (RAPD) techniques.

The date palm is a diploid plant with a complex genome, making it challenging to analyze its genetic diversity. Traditional breeding methods have been used to improve the crop's quality and yield, but these methods have limitations, such as the need for extensive time and resources. Molecular markers, such as RAPD, have been developed to analyze genetic diversity in plants. RAPD is a PCR-based technique that uses short primers to amplify DNA fragments, which are then analyzed to determine genetic diversity.

This study was conducted at the Molecular Genetic Laboratory, Faculty of Agriculture, University of North Sumatra in Medan, from January to July 2018. Two sources of genetic material were used: eight date leaves from propagation through tissue culture methods and two leaves obtained from seed multiplication. The primary RAPD primers used in this study were OPD-13, OPD-20, and OPO-16. The PCR reaction was performed using a thermal cycler, and the amplified DNA fragments were analyzed using agarose gel electrophoresis.

The results of this study showed a high percentage of polymorphics (74.40%) using primary RAPD primers. This indicates a significant genetic variation between the genotypes studied. The size of the DNA fragments ranged from 214 bp to 1918 bp, showing a fairly extensive genetic diversity. The analysis process was carried out using Darwin 6.0 software, which made it easy to group individuals based on genetic similarities.

The results of the analysis revealed that the ten individual dates that were studied could be divided into three main groups, reflecting the difference in molecular diversity between these genotypes. The total molecular diversity was 83.11%, indicating a high level of genetic diversity. The genetic distance coefficient between these individuals ranged from 0 to 0.6, which indicates variations in the level of genetic closeness.

The results of this study demonstrate the effectiveness of the RAPD method in analyzing genetic diversity in dates. This information not only provides insight into the diversity of dates genetic diversity but also has the potential to improve the breeding and conservation program of dates in the future. Knowledge of genetic diversity is very important for the development of better dates varieties, which can adapt to environmental changes and produce higher harvests.
