Optimizing the Hydrolysis Process with the Enzyme Alpha Amylase for the Production of Maltooligosaccharides from the Orange Sweet Potato Paste in Beta Varieties 1
Introduction
The orange sweet potato paste in Beta varieties 1 is one of the leading commodities in North Sumatra, and optimizing its use is crucial to create food products with good nutritional content. The main focus of this study is to determine the optimal enzyme concentration, process conditions (temperature and reaction time), as well as the best enzymatic hydrolysis method for producing maltooligosaccharides from the orange sweet potato paste in Beta varieties 1. This study uses the response surface methodology-Box Behnken Design (RSM-BBD) with Design Expert V.13 software.
Background
Maltooligosaccharides are complex carbohydrates that can function as prebiotics, support digestive health, and improve the immune system. They are highly potential as raw material for healthier food products. The use of alpha amylase as the main enzyme is very crucial, because this enzyme plays a role in breaking starch into smaller units. With the regulation of the concentration of enzymes, temperatures, and appropriate reaction time, the hydrolysis process can be optimized, which in turn increases the efficiency of production and quality of maltooligosaccharides produced.
Methodology
There are three factors studied in this study, namely the enzyme concentration (1-2 U/g), the reaction temperature (50-60 ° C), and the reaction time (30-60 minutes). The results showed that the enzyme concentration factor had a significant effect (p <0.05) on the response of viscosity, total sugar, glucose, reduction sugar, and degrees of polymerization. In addition, the reaction temperature also shows a significant effect (p <0.05) on color responses, viscosity, reduction sugar, beta carotene, and antioxidant content. The reaction time also has a significant impact (p <0.05) on total sugar, glucose, reduction sugar, and antioxidants.
Results
The optimization of the response variable produces a new formula with an enzyme concentration 2 U/g, the reaction temperature is 53.49 ° C, and the reaction time is 40.93 minutes. These results indicate that by manipulating these three factors, we can increase the efficiency of the production of maltooligosaccharides that are highly potential as raw material for healthier food products.

Increasing the use of orange sweet potatoes in beta 1 varieties as a source of maltooligosaccharides can not only increase the nutritional value of food products, but also provide economic added value for farmers and the food processing industry.
